艾美捷PEPC丨磷酸烯醇丙酮酸羧化酶 應用實例:

10µg從擬南芥wt葉組織(Atn非衰老葉)、擬南芥wt葉組織(Ats衰老葉)、紅松針葉組織(PS0、PS36)中用1 M Tris HCl、pH 6.8、10%SDS、15%蔗糖新鮮提取的總蛋白,在10%Bis-Tris Nupage Novex凝膠(120 V/45 min,使用MES緩沖系統)上分離0.5 DTT和在75°C下變性5 min,并在30 min內用聚偏氟乙烯(PVDF)吸附。攪拌45分鐘/RT,用5%脫脂牛奶阻斷印跡。在含有2%脫脂乳的TBS中攪拌,或在/4°C下攪拌,以1:1000的稀釋度在一級抗體中孵育1h/RT。傾析抗體溶液,在RT條件下攪拌,在TBS中短暫沖洗印跡兩次,持續10分鐘。在Agrisera匹配二級抗體(抗兔IgG-辣根過氧化物酶結合物,AS09 602)中培養印跡,稀釋至1:75 000 in,攪拌1h/RT。如上所述清洗印跡,并使用化學發光檢測進行顯影。曝光時間為40秒。
